Histone Arg modifications by PAD4 in cancer and innate immunity

Our lab studies histone modifications in chromatin structure and function. Recently, we found that the peptidylarginine deiminase 4 (PAD4) works as a transcriptional corepressor for the expression of p53-target genes, such as p21 and OKL38. By designing inhibitors to target PAD4, we hope to offer new therapeutic treatments of cancers. Moreover, PAD4 functions as an innate immune factor in neutrophils during bacterial infection. Histone Arg citrullination catalyzed by PAD4 facilitates the release of chromatin to extracellular space to form a structure termed neutrophil extracellular traps that trap and kill bacteria. Our research thus implicates PAD4 as a good cop in fighting bacteria but a bad cop in tumorigenesis by repressing tumor suppressor gene expression.
